Cagrilintide

Description

Cagrilintide is a novel, long-acting acylated analogue of Amylin (islet amyloid polypeptide). Amylin is a peptide hormone naturally co-secreted with insulin by the pancreatic beta-cells.

While GLP-1 agonists (like Semaglutide) focus on the “incretin” pathway, Cagrilintide targets a completely different mechanism: the Amylin/Calcitonin receptor system. In research models, it permeates the blood-brain barrier to act specifically on the satiety centers of the brainstem (area postrema). It is currently the subject of intense investigation as the perfect “partner” to GLP-1s, with studies suggesting it induces a profound sense of “fullness” and suppression of food-seeking behavior that is distinct from, and additive to, the effects of GLP-1 agonists.

• The “CagriSema” Synergy: The most significant application of Cagrilintide is in combination research with Semaglutide (a pairing often referred to in literature as “CagriSema”). Clinical data published in The Lancet suggests that co-administration of these two agents yields weight loss results superior to Tirzepatide, largely because they attack obesity from two independent physiological angles: GLP-1 (metabolic/gastric) and Amylin (neurological satiety).

• Plateau Breaking: Cagrilintide is heavily researched as a “second-line” agent. In subjects who have developed a tolerance or reached a weight-loss plateau on GLP-1 mono-therapy, the introduction of Cagrilintide has been observed to re-sensitize the weight loss trajectory by activating a non-competing pathway.

• Glycemic Control: As an amylin analogue, it plays a role in slowing gastric emptying and suppressing postprandial glucagon secretion, which helps smooth out blood sugar spikes after meals without carrying a high risk of hypoglycemia.
